King of Prussia’s Phio Pharmaceuticals Wins Patent Protection for a New Cancer Drug Candidate Targeting BRD4

Phio Pharmaceuticals, the King of Prussia, Pennsylvania-based clinical-stage biopharmaceutical company, has secured a U.S. Notice of Allowance for a patent covering PH-894, an INTASYL compound designed to selectively silence the BRD4 protein. A Notice of Allowance means the U.S. Patent and Trademark Office has determined the application meets the requirements for a patent and will issue it once final fees are paid, effectively locking in intellectual property protection before the compound has even entered clinical trials.

Why BRD4 Is a Meaningful Target

BRD4 is a well-established target in cancer research, a bromodomain-containing protein that regulates gene expression and has been studied for years as a driver of tumor growth across multiple cancer types, work reflected in a substantial body of independent, peer-reviewed literature on BRD4-targeted therapeutics. Phio describes BRD4 in its own release as “a key regulator of gene expression associated with proliferative and infectious disease,” and PH-894 is designed to use the company’s INTASYL platform to silence it selectively. President and CEO Robert Bitterman framed the patent as part of a broader intellectual property strategy rather than a one-off filing: “Robust intellectual property protection is a continuous focus in our development strategy to advance novel immuno-oncology therapies and maximize the long-term value of the INTASYL platform. This patent advancement solidifies our commitment to advance PH-894 in the U.S. and pursue strategic collaborations internationally.”

How INTASYL Actually Works, and What Else It’s Already Doing

INTASYL is Phio’s proprietary siRNA gene-silencing chemistry, designed to enhance the ability of a patient’s own immune cells to kill cancer cells by shutting down specific genes that would otherwise let tumors evade the immune system. PH-894 isn’t the platform’s first compound to reach a clinical or near-clinical milestone.Phio’s lead program, PH-762, silences the PD-1 gene and completed its Phase 1b trial for cutaneous squamous cell carcinoma, melanoma, and Merkel cell carcinoma in March 2026; the company is now preparing final study documentation for the FDA and intends to request an FDA meeting in the third quarter of 2026 to discuss next steps for the program, giving it a working clinical track record for the same underlying chemistry now being extended to a BRD4-targeted compound. . With this allowance, Phio says it holds 54 issued patents covering INTASYL chemistry, gene targets, and therapeutic applications across major global markets.
