Quadplex dPCR as a tool to analyze genome integrity and heterogeneity in gene therapy drug products

As the gene therapy field rapidly grows, the need for product-specific analytics becomes crucial. Traditional quality attributes often fail to correlate consistently with drug potency or safety. It is believed that the heterogeneity of packaged genomes plays a role in this inconsistency. To address this, a quadruplex dPCR assay using four primer probe sets that cover the genome from promoter to poly(A) signal sequence was developed. This innovative genome integrity analysis allows for the precise calculation of both the absolute number and distribution (heterogeneity) of AAV genome variants. Complementary techniques such as long-read next generation sequencing (NGS) and analytical ultracentrifugation (AUC) can be used alongside this method to provide a thorough characterization of AAV drug products.
